PTPN6 belongs to the class I cysteine based cytosolic tyrosine protein phosphatases. It contains two Src homology (SH2) domains at the N-terminus, and one protein-tyrosine phosphatase domain at the C-terminus. It is expressed predominantly in hematopoietic cells, as well as in nervous system and in epithelial cells of the prostate.

PTPN6 is involved in trophic factor signalling, cell growth and differentiation, chemotactic responses, oxidative stress and tissue inflammation.
