Pannexin-1 is homologous to connexins; it is thought to form plasma membrane hemichannels, but not gap junctions (due to N-glycosylation in the extracellular domain preventing self-association).

Pannexin-1 is thought to be one route by which large fluorescent dye molecules (such as ethidium bromide) may enter cells following P2X7 activation. Proposed functional roles for pannexins are as Ca2+-leak channels to regulate ER Ca2+ homeostasis, and as ATP-release channels.
