MyD88 is an adapter protein containing an N-terminal death domain (DD) and a C-terminal Toll/Interleukin-1 receptor (TIR) domain.

MyD88 mediates signalling pathways initiated by Toll-like receptors (TLRs), IL-1 and IL-18 receptors, and therefore plays important roles in innate and adaptive immunity.

MyD88 interacts with IL-1 receptor-associated kinases (IRAKs) and TNF receptor-associated factor 6 (TRAF6), leading to activation of several transcription factors (e.g. nuclear factor kappa B (NFkB), activator protein 1 (AP-1) and interferon regulatory factors (IRFs)), and induction of cytokine production and immune responses.
